Einen regulären Linienbetrieb nahm die ARCO erst 1964 auf nach Übernehme der Cia. Aeronáutica Uruguaya S.A.., der vier Maschinen vom Typ CurtissC-46 Commando gehörten. Die einzige Route war nur 56 km lang: Sie verband den Flughafen Laguna de los Patos in Colonia del Sacramento in Uruguay am nördlichen Ufer des gewaltigen Mündungstrichters des Rio de la Plata mit dem Aeroparque Jorge Newbery in Buenos Aires am Südufer.
